Rute Q44 dimulai dari persimpangan Jalan Raya Merrick dan Archer Avenue di daerah pusat Jamaica. Kemudian ke arah barat di Archer Avenue, melalui pusat Stasiun Jamaica yang terdiri dari stasiun kereta bawah tanah Archer Avenue dan terminal busnya. Kemudian di stasiun kereta bawah tanah Jalan Raya Sutphin, yang terhubung dengan staisun Jamaica melalui [[Long Island Rail Road]] dan AirTrain JFK, Q44 berbelok ke utara ke arah Jalan Raya Sutphin. Kemudian menuju ke barat ke arah Hillside Avenue dan ke selatan ke arah Jalan Raya Queens. Kemudian di Jalan Utama Queens, Q44 melaju melalui jalanan antara Jalan Raya Queens dan Jalan Raya Utara di Downtown Flushing. Pada tanggal 2 Mei 1933, Perusahaan North Short Bus mulai membuka pelayanan di Jalan Utama antara Jalan Utama dan stasiun kereta bawah tanah Roosevelt Avenue di Flushing serta Jalan Raya Horace Harding (sekarang adalah Jalan Tol Long Islands) di Queensboro Hill. Pada tanggal 18 September 2017, sebuah kecelakaan terjadi antara sebuah bus Q20 dan sebuah bus ''charter'' pribadi dari Dahlia Travel and Tours, dimana bus ''charter'' pribadi tersebut menabrak bus Q20 yang membawa 15 orang penumpang. Tabrakan tersebut membuat bus Q20 berputar dan menabrak Kennedy Fried Chicken yang ada di ujung jalan, menyebabkan kebakaran kecil yang langsung segera dipadamkan.
