Ali Alatas merupakan keturunan blasteran dari [[Bangsa Arab|Arab]] [[Hadhrami]] ([[Yaman]]) dan [[Suku Sunda|Sunda]]. Ia memiliki kakek yang merupakan pedagang pada era Hindia Belanda, yakni [[Abdullah bin Alwi Alatas]]. Alex, begitu ia akrab dipanggil, menikah dengan Junisa dan pasangan ini dikaruniai tiga orang anak. Sebagai diplomat, ia dikenal akrab kepada semua kalangan, baik pejabat maupun petugas keamanan. Ia dilaporkan biasa mengobrol dengan petugas keamanan di PBB sewaktu merokok di luar gedung.
Ia wafat di [[Rumah Sakit Mount Elizabeth]] di [[Singapura]] pada tanggal [[11 Desember]] [[2008]] pukul 07.30 waktu setempat setelah mendapat serangan jantung pada tanggal [[20 November]] [[2008]]. Ia dibawa ke Singapura setelah beberapa hari dirawat di Jakarta.
