Penemuan metamfetamina berawal pada tahun 1871, ketika seorang ahli farmasi Jepang bernama Nagai Nagayoshi yang sedang melakukan riset di [[Humboldt Universität zu Berlin|Universitas Humboldt]], Berlin. Nagoyashi berhasil mengisolasi senyawa efedrina yang berfungsi sebagai stimulan dari tumbuhan Cina, ''Ephedra sinica''. Awalnya efedrina diharpkan dapat membantu penderita asma, namun perusahaan Jerman, Merck, menolak untuk memproduksi obat tersebut karena efeknya yang tidak jauh berbeda dengan adrenalin. Hal ini memicu Nagayoshi untuk meningkatkan efek efedrina dan mengembangkannya menjadi metamfetamina. Sayangnya, Nagoyashi belum dapat menemukan aplikasi praktis metamfetamina dan obat ini akhirnya sempat dilupakan.
