Kuinidin bekerja sebagai penghambat saluran natrium berpagar tegangan.[13][14] Penghambatan saluran Nav1.5 secara khusus terlibat dalam efek antiaritmianya sebagai agen antiaritmia kelas I.[15] Kuinidin juga menghambat [[saluran kalium]] berpagar tegangan tertentu (misalnya, Kv1.4, Kv4.2, hERG, dan lain-lain),[16][17] bekerja sebagai antimuskarinik dan penghambat alfa-1,[18] dan juga merupakan antimalaria.[15] Kuinidin juga dikatakan sebagai antagonis reseptor asetilkolin M3 muskarinik selektif.[19]
