Mekanisme selektivitas saluran kalium tetap menjadi bahan perdebatan yang terus berlanjut. Oksigen karbonil sangat elektronegatif dan penarik kation. Filter ini dapat memuat ion kalium pada 4 situs yang biasanya diberi nama S1 hingga S4 dimulai dari sisi ekstraseluler. Sebagai tambahan, satu ion dapat berikatan di dalam rongga pada situs yang disebut SC, atau satu/lebih ion di sisi ekstraseluler pada situs yang lebih didefinisikan dengan baik disebut S0 atau situs yang kurang didefinisikan dengan baik disebut Sext. Berbagai penempatan berbeda situs tersebut mungkin terjadi. Karena struktur sinar-x rata-rata mencakup banyak molekul, tidak mungkin disimpulkan penempatan sebenarnya secara langsung dari struktur tersebut.
