Penaklukan Honduras oleh Spanyol adalah sebuah konflik abad ke-16 pada kolonisasi Amerika oleh Spanyol dimana wilayah yang sekarang meliputi Republik Honduras, salah satu dari lima negara di Amerika Tengah, dimasukkan ke Kekaisaran Spanyol. Pada 1502, wilayah tersebut diklaim untuk raja Spanyol oleh Christopher Columbus pada perjalanan keempat dan terakhirnya ke Dunia Baru.
